Daytime Lipid Metabolism Modulated by CLOCK Gene Is Linked to Retinal Ganglion Cells Damage in Glaucoma

چکیده

Lipid metabolism is intimately linked to circadian mechanisms and light signaling. Deteriorated photic transduction because of retinal ganglion cell (RGC) loss occurring with glaucoma progression reduces perceived amplitude, causing disruption. To investigate associations RGCs, total cholesterol (TC), its low-density (LDL-C) high-density (HDL-C) fractions, triglycerides (TG) were measured, under a controlled meal regimen, during daytime hours in 114 patients diagnosed primary open-angle (POAG). RGC damage was assessed by high-definition optical coherence tomography (HD-OCT). Analysis eight clock, clock-related, melatonin receptor gene polymorphisms performed on 19 patients. associated changes lipid time-dependent manner. Morning (08:00) values HDL-C (r = 0.613, p < 0.0001) TG 0.568, correlated positively global loss, while LDL-C at 08:00 had weak correlation 0.235; 0.012) but showed strong the evening (20:00) 0.533, 0.0001). The morning–evening gradients (MEGs, 20:00 versus 08:00) TC changed sign from negative positive association exceeding 15% two-eye mean GLV threshold. MEG (LDL-C higher than morning) only POAG CLOCK_3111 TT genotype.

Stem cells, retinal ganglion cells and glaucoma.

Retinal ganglion cells (RGCs) represent an essential neuronal cell type for vision. These cells receive inputs from light-sensing photoreceptors via retinal interneurons and then relay these signals to the brain for further processing. RGC diseases that result in cell death, e.g. glaucoma, often lead to permanent damage since mammalian nerves do not regenerate. Stem cell differentiation can gen...

Manipulating Glia to Protect Retinal Ganglion Cells in Glaucoma

Increasing evidence supports both direct and indirect roles for retinal glia in the pathogenesis of glaucoma. To complicate these roles is the realization that glial activity can be both beneficial and detrimental to the survival of retinal ganglion cells (RGCs) and their axons. The contribution of glia to glaucoma pathogenesis also varies by compartment; glia in retina react differently to dis...

Phototransduction by retinal ganglion cells that set the circadian clock.

Light synchronizes mammalian circadian rhythms with environmental time by modulating retinal input to the circadian pacemaker-the suprachiasmatic nucleus (SCN) of the hypothalamus. Such photic entrainment requires neither rods nor cones, the only known retinal photoreceptors. Here, we show that retinal ganglion cells innervating the SCN are intrinsically photosensitive. Unlike other ganglion ce...

Selective loss of retinal ganglion cells in albino avian glaucoma.

Retinal ganglion cell loss was investigated in the retinae of albino quails before and after the development of glaucoma. The isodensity maps of ganglion cells, the total number of ganglion cells, and the histograms of the cell size in the central region of the retina were similar between albino quails without glaucoma and pigmented quails. However, ganglion cells in the intermediate and periph...

Stem ‍Cells in Glaucoma Management

Glaucoma is the leading cause of preventable blindness worldwide. Despite tremendous advances in medical and surgical management of glaucoma in the recent years, the prevalence of glaucoma related blindness is anticipated to increase in the future decades because of the aging population. Stem cells have the potential to change the glaucoma management in several ways. There are several areas of ...
